Proteome analysis of bovine serum

Abstract
Proteomics is a powerful tool for biomedical investigation as it allows a thorough examination of the qualitative and quantitative composition of a biological sample; comparison between protein patterns, e.g. in diseased versus control status, is then useful to the identification of disease markers.
Definition of a reference map of Bos taurus serum proteins under normal conditions is a first step of an examination of bovine specimens under a variety of test conditions, in order to establish a database of proteomic data. Serum from healthy cows and from animals with udder edema are analysed by two-dimensional electrophoresis and proteins revealed by silver stain. Protein spots will be excised and digested with trypsin in provision for MS analysis and the mass of the resulting peptides evaluated by MALDI. Resulting peptides will eventually be sequenced by ESI. Data from peptide mass fingerprinting and internal sequencing will then be searched against protein databases for protein identification.
