In the recent studies, it was believed that extracts of Cordyceps sinensis and
Cordycepin (3'-deoxyadenosine) have anticancer activity, which was based on
their antiproliferative and cytotoxic effect. Extracts of Cordyceps sinensis and
Cordycepin have already been tested on many different cancer cell lines (mouse
melanoma cell line, mouse lung carcinoma cell line and human oral squamous
cancer cell line); it showed that extracts of Cordyceps sinensis and Cordycepin
can induce programmed cell death (Apoptosis). Therefore it is hypothesized that
such antitumor effects do exist also to neural tumors. To prove this hypothesis
these compounds were be added to well characterize NB cell line (N18TG2) and
their cytostatic effects were be tested dose/time dependently.
In this study, an attempt was made to elucidate a water extract of Cordyceps
sinensis mycelium, on the growth curves of N18TG2 cells (with and without
treatment with water extract of Cordyceps sinensis mycelium). MTT-assay and
PI staining were used to investigate the effect of Cordyceps sinensis water
extract and Cordycepin on cell viability in N18TG2 cells. Data was presented as
concentration and time dependent inhibition curves with statistic calculations.
By the cell culture, we have seen the morphological changes of cells like
rounded-up phenomenon, floated cells, membrane blebbing and cell death with
the treatment of Cordycepin at 1 mM but we did not see these morphological
changes by the treatment with Cordyceps sinensis mycelium water extract.
MTT assay was used to investigate the effect of Cordyceps sinensis water
extract and Cordycepin on cell viability in N18TG2 cells. Results showed that
treatment with Cordyceps sinensis mycelium water extract at the final
concentration of 2.3 mg/ml and 0.23 mg/ml had significantly decrease in cell
number of 38% and 16%. Treatment with Cordycepin at the final concentration of (1 mM, 0.1mM and 0.01 mM) had significantly negative effect on cell
viability in N18TG2 cells, significantly decrease in cell number of 85%, 72%
and 56% (Fig.22 and Fig.23, * p<0.05).
PI staining showed also that Cordycepin at the final concentration of (0.01mM,
0.1mM and 1mM) had significantly cytotoxic effect on neuronal tumor cells.
So it can be concluded that Cordycepin pure has cytotoxic effect at 1 mM, 0.1
mM and 0.01 mM but Cordyceps sinensis water extract have only inhibition
effect on the N18TG2 cell proliferation (cytostatic effect).
