The use of genetic tools in grapevine breeding is getting more important and accelerates the gain of new varieties. Early selection of desired traits in juvenile stage is rendered possible. An open task in viticulture is the nutrition with Mg. Lack of Mg occurs to several cultivars especially on light and acid soils. Welschriesling is one of these cultivars tending to low and inefficient Mg uptake. By means of an interspecific cross of Welschriesling x Sirius, a segregating population was gained. Several offsprings show symptoms of Mg deficiency. The segregation pattern of this defect was compared to the segregation pattern of genetic markers. The double pseudotest-cross mapping strategy was followed by applying 237 SSR (simple sequence repeats) and 14 RAPD (random amplified polymorphic DNA) markers. The SSR markers were involved into the calculations of the genetic way as dominant and codominant alleles. The markers were assembled into 20 linkage groups. The female (Welschriesling) and male (Sirius) map were aligned in one consensus map with Mapmaker 3.0. The map served as the basis of QTL (quantitative trait loci) analysis. Successfully a QTL loci was detected that could be associated with visual symptoms and the amount of Mg-concentrations in leaves of the grape zone. The trait is influenced by a QTL on group 11. The SSR markers VVS 2 and ISV 4 are most close to that trait representing the core region of the QTL.
